Manipulation of Biological Cells with Light

Light is a special form of energy. It has several unique properties that can provide novel manipulation functions of biological cells from cell trapping, sorting, and surgery. This presentation will cover several optical manipulation technologies developed in our group in the past decade. First topic will focus on a laser cell surgery method called photothermal nanoblade that can realize efficient large-sized cargo delivery into fragile mammalian cells. Cargo up to few microns in size can be reproducibly delivered into 100,000 cells on the platform in a minute. The second topic will focus on a high-speed pulse laser activated cell sorter called PLACS that utilizes the ultrahigh energy density feature of pulse laser for rapid actuation of fluid in a highly viscous microfluidic environment. PLACS is the first microfluidic FACS that can realize a throughput higher than 10,000 cells/sec to bridge the gap between micro-fluidic FACS and conventional aerosol FACS. The third topic will cover a novel cell force sensor that utilizes the strong scattering signals from plasmonic gold nanoparticles embedded in elastic micropillar array for massively parallel and precision cell force measurement across a large field of view.

Biography:

Dr. Pei-Yu Chiou received his Ph.D. degree in the Electrical Engineering and Computer Sciences Department from the University of California at Berkeley in 2005. He received his M.S. degree in the Electrical Engineering Depart-ment from UCLA and B.S. degree in the Mechanical Engineering Department from National Taiwan University in 1998. He was an assistant professor in the Mechanical and Aerospace Engineering Department at the University of California at Los Angeles (UCLA) between 2006~2011, associate professor between 2011~2015, and full profes-sor since then. His research interests focus on optofluidics, biophotonics, Lab on Chip, and flexible devices. He received the NSF CAREER award in 2008, JALA TOP TEN innovation award in 2015, UCLA MAE Teaching Award in 2014. He is elected to the fellowship of the College of Fellows at the American Institute for Medical and Biological Engineering (AIMBE) in 2017.
